The authors reviewed experimental studies from their laboratory evaluating adenosine signaling in cats and rats during prolonged wakefulness. Interventions included microdialysis perfusion of adenosine and selective A1 receptor agonists and antagonists into the basal forebrain, single-unit electrophysiological recording of wake-active neurons, and molecular assays (gel shift assays, Western blotting, and mRNA analyses) assessing A1 receptor expression and NF-kappaB DNA-binding activity following 3 to 6 hours of sleep deprivation.
What was found
The abstract reports no numerical values, statistical tests, or effect sizes. Extracellular adenosine selectively increased in the basal forebrain during prolonged wakefulness. Microdialysis perfusion of adenosine or the A1 agonist cyclohexyladenosine increased sleepiness, decreased wakefulness, and inhibited wake-active basal forebrain neurons, whereas the A1 antagonist cyclopentyl-1,3-dimethylxanthine increased wakefulness. Basal forebrain A1 receptor mRNA increased after 6 hours of sleep deprivation. NF-kappaB DNA-binding activity increased after 3 hours of sleep deprivation, accompanied by increased nuclear NF-kappaB protein and loss of cytoplasmic I-kappaB.
Why it matters
These findings identify a positive feedback mechanism wherein prolonged wakefulness elevates basal forebrain adenosine, driving NF-kappaB-mediated upregulation of A1 receptors to mediate homeostatic sleep pressure and sleep debt.
