What was done
Old rats were fed acetyl-L-carnitine (ALCAR; 0.5% or 0.2% wt/vol in drinking water), R-alpha-lipoic acid (LA; 0.2% or 0.1% wt/wt in diet), or both. Cognitive function was evaluated using the Morris water maze for spatial memory and the peak procedure for temporal memory. Hippocampal tissue was examined via immunohistochemistry for nucleic acid oxidation markers (8-hydroxyguanosine and 8-hydroxy-2'-deoxyguanosine) and via electron microscopy for mitochondrial structural changes.
What was found
Supplementation with ALCAR, LA, or their combination improved memory in aged rats. The combined regimen was the most effective across two spatial memory tests (P < 0.05 and P < 0.01) and one temporal memory test (P < 0.05). Age-related oxidative damage occurred predominantly in hippocampal RNA rather than DNA; dietary ALCAR and/or LA significantly reduced oxidized RNA, with the combination showing maximal reduction. Electron microscopy demonstrated that ALCAR and/or LA reversed age-associated mitochondrial structural decay in the hippocampus. Exact numerical test scores, group sizes, and baseline-versus-endpoint quantification were not reported in the abstract.
Why it matters
The study provides mechanistic evidence that co-administering mitochondrial cofactors can mitigate age-associated oxidative RNA damage and mitochondrial structural degradation while partially restoring cognitive performance in aged rodents.
Limits
This is an animal study in rats, precluding direct extrapolation of efficacy, safety, or dosage to humans. The abstract does not report the total sample size (n per group), baseline cognitive scores, treatment duration, or precise quantitative effect sizes for immunohistochemical and ultrastructural analyses.
