What was done
Seventeen healthy male volunteers underwent testing for ventilatory responses to hyperoxic progressive hypercapnia and isocapnic progressive hypoxia across three separate days in a randomized, double-blind design. Participants received intravenous pre-injections of placebo, pirenzepine (a peripheral M1 muscarinic antagonist), or biperiden (a centrally penetrating M1 muscarinic antagonist) before ventilatory response measurements.
What was found
The abstract reports no raw numerical values or effect sizes. There were no significant differences in the mean magnitude of ventilatory responses across the three study days. However, subjects with higher baseline hypercapnic ventilatory responses during placebo testing exhibited significantly greater reductions in response after biperiden administration. This relationship was specific to biperiden and hypercapnia, and was not observed with pirenzepine or hypoxia.
Why it matters
This study provides evidence that central M1 muscarinic receptors are involved in CO2 chemoreception and may explain individual variations in ventilatory sensitivity to hypercapnia.
