What was done

Researchers created nonsuture-associated parietal defects (3, 4, or 5 mm in diameter) in juvenile (6-day-old, n = 15) and adult (60-day-old, n = 15) mice. Calvariae were harvested after 8 weeks and evaluated radiographically and histologically, with percentage healing quantified using Scion Image software analysis of radiographs.

What was found

Juvenile mice healed a significantly greater percentage of their calvarial defects than adult mice across all defect sizes (p < 0.05). Juvenile mean healing was 59% for 3-mm defects, 65% for 4-mm defects, and 44% for 5-mm defects. Adult mice achieved <5% mean healing across all defect sizes, establishing all three sizes as critical-sized defects in adults.

Why it matters

This study establishes a mouse model of age-dependent calvarial bone regeneration, facilitating future mechanistic research using transgenic and knockout strains to study membranous bone healing.
