What was done

In chronically catheterized rats, nutrients were infused into the proximal duodenum or proximal jejunum, and blood was collected via jugular catheters at 0, 30, 60, 90, 120, 180, 240, and 300 minutes to measure ghrelin, insulin, and glucose. Additionally, isocaloric (3 kcal) infusions of glucose, amino acids, or lipids were administered into the stomach or small intestine to evaluate macronutrient-specific effects on circulating ghrelin.

What was found

The depth and duration of ghrelin suppression were equivalent regardless of whether nutrients were infused into the stomach, duodenum, or jejunum. Glucose and amino acids suppressed ghrelin more rapidly and deeply (by approximately 70%) than lipids (by approximately 50%).

Why it matters

These findings indicate that ghrelin suppression does not rely on localized nutrient sensing strictly within the stomach or duodenum. Furthermore, the weaker suppression of ghrelin by lipids provides a potential physiological mechanism for reduced satiety and weight gain on high-fat diets.

Limits

The study was conducted entirely in cannulated rats, limiting direct generalizability to human feeding behavior and physiology. Exact sample sizes and precision estimates (confidence intervals or standard errors) are not reported in the abstract, and downstream effects on long-term caloric intake and body weight were not directly evaluated.
