What was done

Mice subjected to acute ethanol intoxication (4 g/kg i.p.) or chronic ethanol exposure (20% alcohol solution for 13 days) received intraperitoneal injections of BPC 157 (10 mcg/kg), the nitric oxide synthase inhibitor L-NAME (10 mg/kg), and the nitric oxide precursor L-arginine (400 mg/kg), alone or in combination. Treatments were administered 5 minutes before or after acute ethanol, or at 0, 3, or 7 hours into chronic withdrawal. Outcomes assessed included anesthesia duration, righting reflex loss, reaction to external stimuli, hypothermia, mortality, and withdrawal seizures.

Acute ethanol produced sustained anesthesia, complete loss of righting reflex, absent reaction to external stimuli, hypothermia, and 25% mortality. BPC 157 rapidly opposed these acute disturbances and opposed prominent withdrawal seizures. L-NAME aggravated acute intoxication while L-arginine opposed withdrawal. When combined with BPC 157 in acute intoxication, presentations resembled L-NAME treatment but without worsened mortality. In withdrawal, BPC 157 plus L-NAME followed the L-NAME pattern, whereas BPC 157 plus L-arginine mirrored L-arginine.
