Effect of fatty acid chain length on suppression of ghrelin and stimulation of PYY, GLP-2 and PP secretion in healthy men.

What was done
Seven healthy men received 90-minute intraduodenal infusions (rate: 2 ml/min) under three conditions: (1) dodecanoic acid (C12, 0.375 kcal/min), (2) decanoic acid (C10, 0.375 kcal/min), or (3) control. Plasma concentrations of ghrelin, peptide YY (PYY), glucagon-like peptide-2 (GLP-2), and pancreatic polypeptide (PP) were measured during the infusion and following a subsequent buffet meal.
What was found
C12 markedly suppressed plasma ghrelin and increased both PYY and GLP-2 compared with control and C10 (all P < 0.05), whereas C10 had no effect on these hormones. Both C10 and C12 slightly increased PP concentrations (P < 0.05). Absolute hormone values, effect sizes, and buffet intake amounts were not reported in the abstract.
Why it matters
This study shows that the secretion of gut hormones regulating appetite and mucosal function is sensitive to fatty acid chain length, responding to 12-carbon but not 10-carbon fatty acids.
Limits
The sample size was very small (n = 7) and restricted to healthy men. Direct intraduodenal infusion bypasses oral and gastric processing, limiting generalizability to normal dietary intake. Specific numerical values and post-meal outcomes were not provided in the abstract.
