What was done
Healthy men aged 20 to 40 years underwent overnight 12-hour blood sampling at 20-minute intervals to evaluate growth hormone (GH) pulsatility before and 1 week after receiving a single injection of either 60 or 90 µg/kg of CJC-1295, a long-acting GHRH analog. The study evaluated GH pulse dynamics, basal GH secretion, and corresponding changes in circulating IGF-I levels.
What was found
CJC-1295 increased GH secretion while preserving normal GH pulse frequency and pulse magnitude. Basal (trough) GH levels increased 7.5-fold (P < 0.0001), contributing to an overall 46% increase in mean GH levels (P < 0.01) and a 45% increase in IGF-I levels (P < 0.001). No significant differences were observed between the 60 and 90 µg/kg doses. Increases in IGF-I did not correlate with any measured parameters of GH secretion.
Why it matters
This demonstrates that continuous stimulation by a long-acting GHRH analog can elevate sustained trough GH and total IGF-I production without blunting the physiological pulsatility of GH release.
Limits
The abstract does not state the sample size (n). The trial was uncontrolled (no placebo group), tested only a single acute dose, followed subjects for only one week, and was conducted exclusively in young healthy men, limiting generalizability to women, older adults, clinical populations, or chronic use.
