What was done
Middle-aged mice fed a high-calorie diet were administered resveratrol to determine its effects on longevity and metabolic health compared to high-calorie and standard diet controls. The study assessed survival, insulin sensitivity, insulin-like growth factor-1 (IGF-1) levels, AMP-activated protein kinase (AMPK) activity, peroxisome proliferator-activated receptor-gamma coactivator 1alpha (PGC-1alpha) activity, mitochondrial number, motor function, and gene expression changes via parametric analysis of gene set enrichment.
What was found
Resveratrol significantly increased survival and shifted the physiological profile of mice on a high-calorie diet toward mice on a standard diet. Reported physiological improvements included increased insulin sensitivity, reduced IGF-1 levels, increased AMPK and PGC-1alpha activity, increased mitochondrial count, and improved motor function. Gene set enrichment analysis showed resveratrol opposed the effects of the high-calorie diet in 144 out of 153 significantly altered pathways. Exact quantitative values for survival rates, lifespan extension, and metabolic parameters were not reported in the abstract.
Why it matters
This study provided proof-of-concept in a mammalian model that a small molecule can counteract obesity-related pathway dysregulation and extend survival under nutritional stress.
Limits
The experiment was conducted exclusively in mice, so findings cannot be directly applied to human health or longevity. The abstract does not disclose the sample size, dosage of resveratrol, or exact quantitative survival outcomes. The study evaluated middle-aged mice on a high-calorie diet, so findings may not generalize to normal-weight or younger populations.
