What was done
Post-mortem brain sections from individuals with Alzheimer's disease and aged normal controls were analyzed to examine the spatial association between herpes simplex virus type 1 (HSV-1) and amyloid plaques. HSV-1 DNA was identified using in situ polymerase chain reaction (PCR), and amyloid plaques were visualized using immunohistochemistry or thioflavin S staining.
What was found
In Alzheimer's disease brains, 90% of amyloid plaques contained HSV-1 DNA, and 72% of all detected viral DNA was associated with plaques. In aged normal control brains, 80% of plaques contained HSV-1 DNA, but only 24% of the viral DNA was plaque-associated (p < 0.001).
Why it matters
The findings demonstrate direct physical localization of viral DNA within amyloid plaques in human brain tissue, supporting the hypothesis that HSV-1 infection may be involved in amyloid plaque nucleation or pathogenesis.
Limits
The abstract does not disclose the total number of human brain samples analyzed. The study is observational and post-mortem, establishing physical colocalization rather than direct clinical causation, and cannot exclude passive entrapment of viral DNA within pre-existing amyloid deposits.
