What was done
Twenty healthy lean subjects were fasted overnight and given 910 kcal of either a high-fat, high-carbohydrate (HFHC) meal (n = 10) or an American Heart Association (AHA)-recommended meal rich in fruit and fiber (n = 10). Blood was collected before and at 1, 2, and 3 hours postprandially. Measurements included plasma lipopolysaccharide (LPS), LPS-binding protein (LBP), mononuclear cell expression of TLR2, TLR4, and SOCS-3 protein, reactive oxygen species (ROS) generation, and NF-kappaB binding activity.
What was found
The HFHC meal induced significant increases in plasma LPS concentration, mononuclear cell expression of SOCS-3, TLR2, and TLR4 protein, ROS generation, and NF-kappaB binding activity (P < 0.05 for all). These increases were completely absent following the AHA-recommended high-fiber and fruit meal. Exact numerical values and effect magnitudes were not reported in the abstract.
Why it matters
This study shows that macronutrient composition acutely influences postprandial endotoxemia and inflammatory signaling. It provides mechanistic evidence linking high-fat, high-carbohydrate meals to SOCS-3 upregulation, which is implicated in the pathogenesis of insulin and leptin resistance.
