Effect of sodium oxybate on growth hormone secretion in narcolepsy patients and healthy controls.

What was done
Researchers evaluated growth hormone secretion and sleep architecture in 8 male hypocretin-deficient narcolepsy patients with cataplexy and 8 healthy male controls matched for age, BMI, waist-to-hip ratio, and body fat percentage. Both groups underwent 24-hour blood sampling at 10-minute intervals before and on the fifth consecutive night of sodium oxybate administration (3 g taken twice per night). Growth hormone concentration time series were analyzed using AutoDecon and approximate entropy alongside sleep recording.
What was found
At baseline, basal and pulsatile growth hormone secretion, pulse regularity, pulse frequency, and approximate entropy values were similar between narcolepsy patients and controls. Administration of sodium oxybate caused a significant increase in total 24-hour growth hormone secretion rate in narcolepsy patients, but not in controls. Slow-wave sleep and the cross-correlation between growth hormone levels and slow-wave sleep more than doubled in both groups after sodium oxybate administration. Specific numerical values and statistical test values were not reported in the abstract.
Why it matters
The study shows that sodium oxybate enhances slow-wave sleep and increases nocturnal growth hormone secretion in narcolepsy patients while strengthening the temporal link between deep sleep and hormone release. This somatotropic effect provides a physiological mechanism for nonsleep clinical benefits, including body weight reduction.
Limits
The sample size is small with only 16 total male participants, excluding females from the evaluation. The trial lacked a randomized placebo control group, and the intervention was short at only five nights. Additionally, numerical values and variance metrics were not provided in the abstract.
