What was done

Rat pups received intranasal delivery of endothelin receptor antagonists to block local survival factors and induce apoptosis in the olfactory epithelium. Investigators evaluated olfactory responses using electroolfactograms and tested behavioral performance with a maternal odor orientation assay against untreated littermates. To assess activity-dependent rescue, pups were exposed to octanal (a ligand for the rI7/Olr226 receptor) for 10 days, and rI7-expressing olfactory sensory neuron (OSN) abundance was measured by RT-qPCR and whole-mount in situ hybridization.

What was found

Endothelin receptor antagonist treatment increased overall apoptosis in the olfactory epithelium and decreased electroolfactogram responses to odorants. Despite reduced general responsiveness, treated pups performed better in maternal odor orientation tests than controls. Antagonist treatment decreased rI7-expressing OSN counts, but concurrent octanal odorization rescued this loss specifically for rI7 OSNs, with optimal protection at low odorant concentrations. Octanal exposure alone did not alter rI7 OSN numbers in untreated littermates. The abstract reports no exact numerical values or effect sizes.

Why it matters

The findings demonstrate that elevated apoptosis in the olfactory mucosa can accelerate activity-dependent neuronal selection, biasing the sensory neuron population toward actively encountered environmental odors.

This study was conducted exclusively in rat pups, limiting generalizability to adult animals or humans.
