What was done
Elderly volunteers were evaluated to determine whether treatment with the mTOR inhibitor RAD001 could ameliorate immunosenescence. The primary functional outcome was the immune response to influenza vaccination, alongside measurement of programmed death-1 (PD-1) receptor expression on CD4 and CD8 T lymphocytes.
What was found
RAD001 treatment enhanced the immune response to the influenza vaccine by approximately 20% at doses reported to be relatively well tolerated. It also decreased the percentage of CD4 and CD8 T lymphocytes expressing the inhibitory PD-1 receptor.
Why it matters
This study provides clinical trial evidence in humans that pharmacological mTOR inhibition may help reverse aspects of age-related immune decline and improve vaccine responsiveness in older adults.
