What was done
Genotyping of seven FOXO3 polymorphisms was performed in a community-based case-control study of 1336 Southern Chinese individuals living in the Red River Basin. This was combined with a meta-analysis of eight case-control studies comprising 5327 longevity cases and 4608 controls, including ancestry-stratified analyses.
What was found
In the case-control study, rs2802288*A (pallele = 0.005, OR = 1.266) and rs2802292*G (pallele = 0.026, OR = 1.207) were significantly associated with longevity following Bonferroni correction. Among the longevity group, carriers of rs2802288*A and rs2802292*G showed reduced HbA1c (p = 0.001), and rs2802292*GG homozygotes showed improved HOMA-IR (p = 0.014). The meta-analysis confirmed overall contributions of both variants to longevity, with rs2802292*G showing potentially stronger effects in Asians than Europeans. Numeric pooled effect sizes and confidence intervals for the meta-analysis were not reported in the abstract.
Why it matters
This paper extends FOXO3 longevity associations to Southern Chinese in the Red River Basin and links these genetic variants to improved glycemic control and insulin sensitivity in long-lived individuals.
