Evidence for a Long-Lasting Compulsive Alcohol Seeking Phenotype in Rats.

Alcohol-preferring rats were identified using an intermittent two-bottle choice access procedure and subsequently trained on a chained schedule of reinforcement to seek and take alcohol. Following stable acquisition, unpredictable electric foot-shocks (0.45 mA) were introduced during seeking cycles to test for punishment-resistant behavior. Rats were tracked longitudinally to assess the duration of compulsive seeking. Additionally, researchers evaluated the effects of the selective mu-opioid receptor antagonist GSK1521498 on alcohol seeking and consumption in both compulsive and non-compulsive phenotypes.
What was found
Punishment-resistant compulsive alcohol seeking emerged in only a subset of alcohol-preferring rats and persisted for nearly a year. Administration of GSK1521498 reduced both alcohol seeking and alcohol intake in both compulsive and non-compulsive animals. The abstract reports no numerical values or effect sizes.
Why it matters
