Evidence for a role of testosterone-androgen receptor interactions in mediating masculine sexual behavior in male rats.

What was done
Sexually experienced castrated male rats received subcutaneous Silastic capsules delivering physiological doses of testosterone (T), 10% estradiol (E2), dihydrotestosterone (DHT), 10% E2 plus DHT, or blank implants. Copulatory behavior (mounting and ejaculation) was assessed on days 2-4, 5-7, 10-12, and 14-16. Cell nuclear androgen receptor (AR) and estrogen receptor (E2R) binding were measured in the hypothalamus, preoptic area, amygdala, and septum. Additional experiments evaluated whether 0.5% E2 plus DHT (matching T-treated receptor levels) or 1.0% E2 plus DHT (matching intact male E2R levels) could restore sexual behavior.
What was found
T, 10% E2, and 10% E2 plus DHT all activated mounting behavior, but only T restored ejaculation in 100% of castrated males. DHT alone showed no effect on male sexual behavior. When AR and E2R occupancy levels from T-treated males were matched using 0.5% E2 plus DHT, male rats showed virtually no sexual behavior. Similarly, rats exposed to 1.0% E2 plus DHT failed to display male sexual behavior despite matching intact E2R levels. Specific numerical receptor binding values were not provided in the abstract.
Why it matters
The findings demonstrate that testosterone-androgen receptor interactions are essential and distinct in mediating complete masculine sexual behavior, which cannot be fully replicated simply by co-administering DHT and estradiol to match receptor occupancy levels.
Limits
The experiment was conducted in an animal model (rats), limiting direct clinical translation to humans. The abstract does not report group sample sizes (n) or exact numerical behavioral frequencies.
