What was done
Ten rats were subjected to total sleep deprivation (TSD) using the disk apparatus with yoked controls (TSC). Physical appearance, survival time, body weight, food intake, and energy expenditure (calculated from caloric value of food, weight, and waste) were evaluated from the abstract.
What was found
All 10 TSD rats died or were sacrificed when death seemed imminent within 11-32 days, whereas yoked controls remained healthy. No anatomical cause of death was identified. TSD rats showed a debilitated appearance, lesions on tails and paws, and weight loss despite increased food intake. Energy expenditure increased in all TSD rats, with mean levels reaching more than twice baseline values.
Why it matters
This landmark animal study demonstrates that sustained total sleep deprivation is fatal in rodents and induces severe hypermetabolism where energy expenditure exceeds compensatory caloric intake.
Limits
The findings are derived from a small sample of rodents (10 TSD rats) using an experimental apparatus, precluding direct clinical extrapolation to humans. A specific anatomical or physiological mechanism for death was not identified.
