Genome-wide cross-disorder meta-analyses and genetic correlation analyses were performed on aggregate data from 232,964 cases and 494,162 controls across eight psychiatric conditions: anorexia nervosa, attention-deficit/hyperactivity disorder, autism spectrum disorder, bipolar disorder, major depression, obsessive-compulsive disorder, schizophrenia, and Tourette syndrome.

What was found

Genetic correlation analyses identified three clusters of inter-related disorders. Meta-analysis revealed 109 loci associated with at least two psychiatric disorders, including 23 loci showing pleiotropic effects across four or more disorders and 11 loci with antagonistic effects on multiple disorders. Pleiotropic loci were enriched in genes showing heightened brain expression across the lifespan starting prenatally in the second trimester and participating in neurodevelopmental processes.

Why it matters

This study demonstrates that genetic risk factors cross traditional clinical diagnostic boundaries, identifying shared biological pathways and neurodevelopmental timing that could inform psychiatric classification and cross-disorder drug targets.
