What was done
Intact and bilaterally adrenalectomized (ADX) adult male rats maintained on a 14:10 light:dark cycle received 10 IU insulin intraperitoneally at 1300 h. Groups of rats (n = 8) were killed at 2, 3, or 4 hours post-injection alongside uninjected controls. Plasma catecholamines were quantified using high performance liquid chromatography, and pineal N-acetyltransferase (NAT) activity and melatonin content were measured by radioimmunoassay.
What was found
Insulin induced hypoglycemia in all injected animals. In intact rats, plasma epinephrine rose at 2 hours compared to uninjected controls, accompanied by increased pineal NAT activity at 2 hours and significantly elevated melatonin content at 2, 3, and 4 hours. In ADX rats, plasma epinephrine did not rise, and pineal NAT activity and melatonin content showed no changes. Exact numerical values, baseline concentrations, and confidence intervals were not reported in the abstract.
Why it matters
The study indicates that an adrenal factor, likely epinephrine, mediates the stimulatory effect of acute insulin-induced hypoglycemia on pineal melatonin synthesis in rats.
Limits
This is an animal study conducted exclusively in male rats with daytime acute stress, limiting direct clinical translation to humans. Total animal count is unclear beyond the per-timepoint group size (n = 8), and quantitative measurements and variance data are absent from the abstract.
