What was done
The authors evaluated the convergent validity of three dominance markers (urinary darcin, tube test score, and preputial gland-to-body length ratio) against wound severity and home-cage behavioral rankings in group-housed male mice using factor analysis. Discriminant validity between tube test scores and anxiety was assessed via open-field testing. The sample consisted of 24 cages (N = 24) balanced across two strains (SJL and albino C57BL/6) and two group sizes (3 or 5 mice). Home-cage behaviors were recorded in the first week, and experimental dominance metrics were collected in the second week.
What was found
The abstract reports no numerical statistics, correlation coefficients, or p-values. After controlling for strain and group size, darcin and preputial ratio had strong factor loadings on the same factor, which significantly predicted home-cage dominance ranking. Tube test scores were not significantly affected by open-field measures. Social network analysis showed that despotic hierarchies predominated, dominant aggressors were more active and rested away from cage mates, and individual social investigation and aggression rates were highly correlated.
Why it matters
Biochemical and anatomical markers such as urinary darcin and preputial gland ratios can identify dominant male mice without labor-intensive behavioral observation, facilitating targeted aggression management in laboratory housing.
Limits
