This narrative review synthesized literature spanning over 25 years on the antioxidant and anti-inflammatory activities of gamma-tocopherol (γT) and its metabolites relative to alpha-tocopherol (αT). It examined in vitro mechanistic data, animal disease models of asthma, cancer, and inflammation, and clinical studies in patients with kidney disease and mild asthma.

What was found

The abstract reports directional and mechanistic findings without quantitative numbers. γT, unlike αT, traps reactive nitrogen species by forming 5-nitro-γT, blocks 5-lipoxygenase translocation in leukocytes to inhibit leukotrienes, decreases cyclooxygenase-2-mediated prostaglandins in macrophages, and inhibits cancer cell growth while sparing healthy cells. Its metabolites, 13'-carboxychromanol and carboxyethyl-hydroxychroman, exhibit COX-1/COX-2/5-LOX inhibition and natriuretic activity, respectively. Preclinical supplementation mitigated disease symptoms in inflammation, asthma, and cancer models, and clinical supplementation reduced inflammatory markers in kidney disease and mild asthma.
