What was done
Realtime 2D cell culture analyses were used to examine lipid uptake, lipid droplet size, lipolytic activity, and gene expression in adipocytes. The cultures were exposed to oleic acid under high glucose (4.5 g/L) versus low glucose (1 g/L) conditions, with and without insulin treatment.
What was found
The abstract reports no numerical values or effect sizes. Qualitatively, high glucose was required for oleic acid to enlarge lipid droplets into unilocular-like mature adipocyte morphology. Oleic acid exposure reduced PPARG gene transcription. Insulin prevented this oleic acid-induced increase in lipid droplet size and inhibited the transcription of lipid-storage genes CIDEC and G0S2, without altering lipolytic activity between high and low glucose states.
Why it matters
The findings demonstrate that insulin can play a protective role against excessive fatty acid-induced adipocyte hypertrophy in vitro by modulating specific lipid-storage gene expression.
Limits
