What was done

The authors evaluated the in vitro anthelmintic activity of eugenol against the adult and muscle larvae stages of *Trichinella spiralis*. Parasites cultured in RPMI-1640 medium were exposed to five eugenol concentrations (122, 305, 609, 1218, and 3045 µM) alongside negative (untreated) and positive controls (albendazole, 377 µM in triplicates). Parasite mortality was tracked at 1, 3, 6, 10, and 24 hours, and structural cuticular alterations were examined via scanning electron microscopy (SEM). Half-maximal inhibitory concentrations (IC50) were calculated.

What was found

The calculated IC50 for eugenol was 187.5 µM for muscle larvae and 190.4 µM for adults. Eugenol demonstrated time- and concentration-dependent mortality, with adult stages being less susceptible than muscle larvae. Eugenol achieved 100% mortality against muscle larvae after 10 and 24 hours at concentrations of 1218 and 3045 µM, equivalent to the albendazole positive control. For adult parasites, concentrations of 1218 and 3045 µM had a significant effect on mortality (p < 0.0001) with a significant concentration-by-time interaction (p < 0.0001). SEM confirmed massive destruction of parasites and complete loss of cuticular striations at 1218 and 3045 µM.

Why it matters

Eugenol shows potent direct in vitro anthelmintic activity against both adult and larval stages of *T. spiralis*, presenting a potential lead candidate for alternative treatments against trichinosis.

Limits

This study was conducted entirely in vitro, without in vivo evaluation of bioavailability, tissue penetration into muscle cysts, host toxicity, or therapeutic index. The exact sample size (total number of worms or larvae tested per condition) was not stated in the abstract.
