What was done

The author conducted a narrative review re-evaluating historical and contemporary literature on mammalian gastrointestinal melatonin synthesis. The review examined the origin of the claim that the gut contains 400-fold more melatonin than the pineal gland, evaluated assay cross-reactivity in tryptophan-loading experiments, and assessed circulating melatonin levels following pinealectomy.

What was found

A 1992 benchmark study reported rat duodenal melatonin at 24 000 ± 2000 pg/g (range: 4000-100 000 pg/g) compared to pineal melatonin at 580 000 ± 36 000 pg/g, which was subsequently miscalculated and cited across literature as evidence that the gut produces 400 times more melatonin than the pineal gland. Re-analysis of subsequent measurements demonstrates that the actual ratio of duodenal to pineal melatonin is likely 0.05-0.19:1. Furthermore, evidence shows pinealectomy eliminates circulating melatonin, whereas contrary findings were driven by cross-reactivity in unvalidated immunoassays with tryptophan and its metabolites.

Why it matters

This analysis challenges a 30-year dogma in pineal and gastrointestinal physiology by demonstrating that daytime circulating melatonin is not gut-derived and that the mammalian gastrointestinal tract is not a major extra-pineal source of the hormone.

Limits

This is an unsystematic narrative review with no formal search protocol, study selection criteria, or quantitative meta-analysis. The underlying evidence rests almost entirely on animal models and bench assay validation rather than direct human physiological data, and no new primary laboratory measurements were conducted.
