In vitro enzymatic assays were conducted to determine whether quercetin-3-glucuronide, a circulating human metabolite of quercetin abundant in red wine, inhibits mitochondrial aldehyde dehydrogenase 2 (ALDH2).
What was found
Quercetin-3-glucuronide inhibited ALDH2 with an IC50 of 9.6 µM, a concentration noted to be comparable to reported circulating levels following red wine consumption.
Why it matters
It provides a testable biochemical hypothesis for red wine-specific headaches, suggesting quercetin metabolites inhibit acetaldehyde metabolism similarly to genetic ALDH2 deficiency.
Limits
This is strictly an in vitro enzyme inhibition study without human clinical or in vivo pharmacokinetic testing. Actual intracellular inhibition in human tissues and causation of headache symptoms were not evaluated.
