Researchers performed 16S rRNA gene sequencing on 30 postmortem brain tissue samples taken from frontal cortex, hippocampus (AD-associated regions), and hypothalamus (non-AD region) across 4 individuals with normal brain histology and 4 Alzheimer's disease (AD) patients, alongside 3 blank controls, to characterize brain bacteriota.

What was found

Brain bacteriota exhibited remarkably low biomass, and overall bacterial community structures were delineated primarily by brain region rather than AD status. Most analyzed parameters showed no significant differences between normal and AD groups. However, *Cloacibacterium normanense* was uniquely detected in AD-associated neurodegenerative regions, and infection-associated bacteria (rather than periodontal pathogens) were enriched in AD brains. No exact numerical metrics or p-values were reported in the abstract.

Why it matters

This study provides evidence on the human brain microbiome in AD, suggesting that while overall bacterial profiles are primarily tissue-specific and low-abundance, selective infection-associated bacteria may correlate with AD neurodegeneration.

Limits

The sample size was extremely small (4 AD patients, 4 controls; 8 individuals total). Low-biomass sequencing is vulnerable to background contamination. The abstract reports no quantitative values or statistical effect sizes, and the postmortem cross-sectional design cannot establish whether bacterial presence drives neuroinflammation or results from blood-brain barrier breakdown.
