What was done
Researchers evaluated CTX130, an allogeneic CD70-targeting CAR T-cell therapy, across preclinical renal cell carcinoma (RCC) xenograft models and in a multicenter, first-in-human phase I clinical trial. The trial enrolled 16 patients with relapsed or refractory advanced clear cell renal cell carcinoma (ccRCC) to assess safety, dose-limiting toxicities, and preliminary efficacy. Preclinical testing of a second-generation edited construct, CTX131, was also conducted.
What was found
In preclinical testing, CTX130 induced complete regression of RCC xenograft tumors. In the phase I trial (n = 16), zero patients experienced dose-limiting toxicity. A disease control rate of 81.3% was achieved, with one patient maintaining a durable complete response at 3 years. Preclinical evaluation of the modified construct CTX131 showed improved expansion and anti-tumor efficacy compared to CTX130.
Why it matters
CAR T-cell therapies have historically shown limited efficacy in solid tumors. These findings provide clinical proof of concept and manageable safety for allogeneic CD70-targeted CAR T-cell therapy in advanced clear cell renal cell carcinoma.
Limits
The clinical cohort was very small (n = 16), non-randomized, and uncontrolled. Survival metrics beyond the single durable responder were not reported in the abstract. Preclinical findings and the modified CTX131 construct have not yet been established in human trials.
