Researchers evaluated the amino acid profiles and protein compositions of myofibrillar and muscle connective protein fractions isolated from skeletal muscle tissue to define their specific constituents.

The muscle connective protein fraction contained ~70% of total mixed muscle collagen, with collagen representing 4.4 ± 0.9% of total protein in that fraction versus 1.2 ± 0.2% in mixed muscle tissue (p < 0.05). Myofibrillar proteins accounted for 39% of the myofibrillar protein fraction and 32% of the muscle connective protein fraction. Key scaffolding proteins comprised 42% of the connective protein fraction compared to 11% of the myofibrillar fraction.

Nutritional interventions often evaluate fraction-specific muscle protein synthesis rates; understanding fraction composition clarifies the degree of collagen enrichment and cross-fraction contamination.
Limits
The abstract does not state the sample size, donor species or characteristics, specific muscle sampled, or fractionation methodology. Incomplete fraction separation was evident, with myofibrillar proteins making up nearly a third of the connective protein fraction.
