What was done

Moringa oleifera seed powder was extracted via microwave-assisted 70% ethanol extraction, purified using macroporous resin, and hydrolyzed with myrosinase. The parent glucosinolate and its enzymatic product were identified as 4-(α-L-rhamnopyranosyloxy) benzyl glucosinolate (4-RBMG) and benzyl isothiocyanate (BITC) using UV-Vis, FT-IR, NMR, and mass spectrometry. In vitro assays were conducted to compare antioxidant potential (DPPH and ABTS radical scavenging at 5–20 mg/mL), anti-inflammatory activity (nitric oxide inhibition), and anti-tumor efficacy across five cell lines, including HL-60 leukemia, A549 lung cancer, and HepG2 hepatocellular carcinoma cells.

What was found

At concentrations of 5–20 mg/mL, 4-RBMG displayed higher DPPH and ABTS radical scavenging activity than BITC. In contrast, BITC exhibited superior anti-inflammatory and anti-tumor activity. At 6.25 μmol/L, BITC inhibited nitric oxide production by 96.67% without observed cytotoxicity. At 40 μmol/L, BITC inhibited growth across all five tested tumor lines, reaching over 90% inhibition in HL-60, A549, and HepG2 lines.

Why it matters

The findings illustrate that enzymatic hydrolysis of Moringa oleifera glucosinolates into isothiocyanates shifts the functional profile from radical scavenging to potent cellular anti-inflammatory and cytotoxic activities in vitro.

Limits

This study is limited entirely to cell-free and cell-culture assays. It does not provide in vivo pharmacokinetics, systemic toxicity profiles, bioavailability metrics, or therapeutic efficacy in whole-organism models. Detailed statistical variance, IC50 metrics, and data on the other two tumor cell lines were omitted from the abstract.
