The review evaluated glycyl-L-histidyl-L-lysine-copper (GHK-Cu) as a standalone intervention in aesthetic medicine across 20 included studies (18 preclinical studies and 2 randomized controlled trials).
What was found
The abstract reports no numerical values, effect sizes, or confidence intervals. Preclinical models demonstrated that GHK-Cu enhanced type I collagen and glycosaminoglycan synthesis, modulated metalloproteinase activity, stimulated angiogenesis and cell proliferation, and decreased TGF-β and IL-6. Clinically, two randomized controlled trials reported reduced wrinkle depth and volume compared to controls and improved patient satisfaction following laser resurfacing.
Why it matters
While GHK-Cu is popular in commercial and clinical aesthetic practices, this review highlights that the vast majority of supporting data is preclinical, emphasizing the lack of standardized clinical protocols.
Limits
Only two human randomized controlled trials were identified, and the abstract provides no human sample sizes, numerical outcomes, or safety and adverse event rates. Formulations, delivery methods, and dosing regimens were heterogeneous across studies.
