Suprachiasmatic nucleus of the human brain: an immunocytochemical and morphometric analysis.

What was done
An immunocytochemical and morphometric analysis was conducted on post-mortem hypothalamic tissue from 42 human autopsy subjects (22 males and 20 females, aged 10 to 92 years). Serial coronal paraffin sections (6 µm) were sampled every 25th section and stained for arginine vasopressin (VP) or vasoactive intestinal polypeptide (VIP) using the peroxidase-antiperoxidase method. Cell counts, nuclear diameters, subdivision volumes, and rostrocaudal lengths were quantified unilaterally under 500x magnification using deconvolution and section thickness corrections, evaluated across sex and age groups.
What was found
The VP subdivision occupied the dorsomedial SCN with a volume of 0.244 ± 0.017 mm3 and contained 6,890 ± 520 neurons (density ~29,000 neurons/mm3). Rostrocaudal length was longer in females than males (1.76 ± 0.12 mm vs. 1.40 ± 0.10 mm), but total VP cell count showed no significant sex- or age-related alterations. The VIP subdivision occupied the ventral and central SCN with a volume of 0.034 ± 0.004 mm3 and contained 1,700 ± 140 neurons (density ~63,000 neurons/mm3), with rostrocaudal lengths of 1.07 ± 0.08 mm in females and 1.02 ± 0.11 mm in males. VIP cell counts showed age-dependent sexual dimorphism: young males had about twice as many VIP neurons as young females, middle-aged females had about 1.7 times as many as middle-aged males, and the difference disappeared in old subjects.
Why it matters
This study establishes quantitative baseline dimensions for peptidergic subpopulations in the human circadian pacemaker, showing VP neurons outnumber VIP neurons roughly fourfold and identifying age-dependent sex differences in VIP neuronal counts.
Limits
The sample size is modest (n = 42) and small when stratified by age and sex subgroups. Measurements reflect peptide immunoreactivity rather than total neuronal populations, which can be influenced by post-mortem factors or peptide expression levels. No direct functional or physiological circadian measures were assessed.
