What was done

The authors reviewed published literature evaluating the nicotine pharmacokinetic profile of JUUL e-cigarettes, as well as laboratory studies quantifying nicotine concentration in JUULpod liquid, aerosol emissions via machine-puffing protocols, and systemic absorption in human users (incorporating 8 pharmacokinetic studies, 5 liquid quantification studies, and 7 aerosol studies).

What was found

Liquid analysis in 5 independent studies showed 39.3 to 48.3 mg nicotine per 5% pod. In 7 vaping-machine studies, nicotine output ranged from 72 to 164 µg per puff, or 14.4 to 32.8 mg per pod at 200 puffs (equivalent to 13–30 cigarettes), with one study finding 68% transfer efficiency (28.8 mg/pod). Across 8 pharmacokinetic studies, peak nicotine levels were half to three-quarters of combustible cigarettes in industry studies with JUUL-naïve users, but equal to or higher than combustible cigarettes in independent studies with experienced users. A 5-day controlled switch study measured nicotine intake equivalent to 18 cigarettes per pod.

Why it matters

This synthesis clarifies the cigarette-pack equivalence of a single JUULpod and shows that while nicotine delivery is lower in initial naïve use, experienced users achieve nicotine absorption equal to or exceeding combustible cigarettes.

Limits

The abstract describes a narrative synthesis of heterogeneous laboratory methods and human trials rather than a systematic meta-analysis. Total human sample size (n) is not reported in the abstract, and results varied significantly based on user experience level and industry versus independent study sponsorship.
